Human Polyomavirus JC (JCV) belongs to the Polyomaviridae family. JCV frequently establishes latent and/or persistent infections, with presence in different organs including the kidney, and remains dormant without causing any disease until the onset of a state of natural or iatrogenic immunosuppression. In these contexts, JCV can reactivate and cause progressive multifocal leukoencephalopathy (PML), a fatal demyelinating disease of the central nervous system, documented in heart, kidney and liver transplant recipients. There is also evidence of a potential role of JCV in the development of brain and colon tumors.

Currently the most sensitive diagnostic technique for the identification of the viral genome is represented by Real-Time PCR. In fact, the diagnosis of JCV on biopsy of the affected organ by electron microscopy and immunohistochemistry is not characterized by suitable sensitivity, and the serological investigation has little value due to the high diffusion of the virus in the human population.
